San Polo dei Cavalieri developed as a medieval stronghold in a strategic position between Roman Sabina, Tivoli and the Aniene Valley. Early documentary traces refer to the fundus Janule and Castrum Sancti Pauli in Jana, while the castle grew as a control point over the valleys below.

Over the centuries the village passed from the Orsini to the Cesi and then the Borghese. Under the Cesi, the castle gradually lost its purely military role and became a noble residence. During the 17th century San Polo was also connected with the history of the Accademia dei Lincei, one of the most prestigious chapters in Italian scientific culture.
